Abstract: The following research work was carried out on the grounds of the agricultural engineering career of the Universidad Laica Eloy Alfaro de Manabí, El Carmen extension, known as "Río Suma", located on the Santo Domingo - El Carmen road, km. 36 on the right margin, with the objective of evaluating two pre-germinative treatments and three substrates in the sexual propagation of guaiacum (Guaiacum officinale). The objective was to evaluate two pregerminative treatments and three substrates in the sexual propagation of guaiacum (Guaiacum officinale); for the development of this research a completely randomized block design (DBCA) was established in factorial arrangement A x B + N; distributed with 7 treatments and 3 replications, the data btained were compared with the tukey test at 5% probability, using the computer program InfoStat. Factor A was formed by the substrates (sawdust, husk and sand) and factor B by the pre-germinative treatments (coconut water and mineral water), the data taken were the germination percentage, leaf diameter and plant height. The results btained determined that the use of substrates and pre-germinative treatments on guaiacum seed did not affect its germination percentage; the average germination percentage was 88.69%; In the case of plant development, the treatments showed statistical differences in the diameter of the plant leaf, as well as differences in the pregerminative treatment factor and the interaction of the factors, with the sawdust substrate and coconut water being the best with an average diameter of 2.46 cm; however, in the height of the plant, none of the factors influenced this parameter.
